Understanding LDH Reagent

Serum lactate dehydrogenase (LD) levels rise due to various conditions such as myocardial infarction, liver disease, anemias, pulmonary emboli, malignancies, and muscular dystrophy. Analyzing LD and CK (creatine kinase) isoenzymes together can confirm acute myocardial infarction.

Methodology

The LD test is based on the method developed by Wacker et al. It involves the conversion of lactate to pyruvate catalyzed by LD, producing NADH as a byproduct. NADH absorbs light strongly at 340 nm, allowing for measurement of LD activity in the sample based on the rate of change in absorbance at this wavelength.
